This is an enzyme-catalyzed reduction by the (S)-selective enzyme Gre2p in phosphate-buffered aqueous solution. The enzyme uses NADPH as a cofactor, which is oxidized to NADP+ in the reaction process. This cofactor was recycled using glucose dehydrogenase. First, a mixture of 100 mM Glucose, 1 mM NADP+, 1.9 µM GDH (glucose 1-dehydrogenase) and 1 mM MgCl2 (co-factor for the GDH) in KPi (KH2PO4/K2HPO4) buffer (100 mM, pH 7.5) was preincubated for approx. 20 minutes for production of NADPH. Then, 0.7 µM of Gre2p and 5-nitrononane-2,8-dione (201 mg, 1.00 mmol, 1.00 equiv) were added. The reaction was shaken at 30 °C for 5 hours.
